fa بررسی ویروس موزاییک یونجه در مناطق شمالی و مرکزی استان خراسان عمومی Ggeneral پژوهشي Research بررسی پراکنش‌ آلودگی ویروس‌ موزاییک(Alfalfa mosaic virus ,ALMV ) با استفاده از آزمون DAS-ELISA نشان داد که مزارع یونجه،سیب زمینی وگوجه فرنگی شهرستان‌های چناران، قوچان، شیروان، مشهد، نیشابور و تربت حیدریه همگی به این ویروس آلوده می‌باشند. با توجه به آزمون کای مربع بین این مناطق از لحاظ میزان آلودگی اختلافی مشاهده نگردید. میانگین کل آلودگی در این مناطق 53 درصد به دست آمد.جدایه جمع آوری شده از مزرعه یونجه مشهد در <em>Nicotiana tabacum</em> L.cv.<em>Samsun</em> تکثیر و از آن برای خالص سازی،تعیین دامنه میزبانی و برخی مطالعات دیگر استفاده شد. مایه زنی مکانیکی این جدایه از ویروس مـــوجب بــروز لـــکه های مـــوضعی در <em>Chenopodium quinoa</em> <em>،C. amaranticolor</em> <em>،Vigna unguiculata</em> ، <em>Phaseolus vulgaris</em> cv.<em>Red kidney</em> و علائم سیستمیک رگبرگ روشنی و موزائیک در <em>Nicotiana glutinosa</em> <em>،N. tabacum</em> cv.<em>Samsun</em> <em>،Ocimum basilicum</em> <em>،Cicer arietinum</em> و <em>Lycopersicon esculentum</em> گردید.در مورد <em>Cucumis sativus</em> علائمی ظاهـــر نشد. خالـــص سازی ویروس ALMV با استفاده از روش ارائه شده توسط کایزر و روبرتسون انجام شد. راندمان خالص سازی ویروس 05/11 میلی گرم ویروس به ازای هر 100 گرم بافت آلوده بود. ویروس خالص شده در برابر جدایه آمریکایی ALMV باند رسوبی تشکیل داد. با انجام الکتروفورز SDS-PAGE و Western blot وزن مولکولی پروتئین پوششی ویروس 24 کیلو دالتون تعیین گردید. از این لحاظ جدایه مورد بررسی با سایر جدایه‌های گزارش شده تفاوت بارزی نشان نداد. The study of dispersion of alfalfa mosaic virus (ALMV) infection based on DAS-ELISA indicated that the fields of Alfalfa, potatoes and tomatoes from Chenaran, Ghochan, Shirvan, Mashhad, Neishaboor and Torbat Heydarieh were infected with the virus. The Statistical analysis indicated that the amount of infection did not differ in the surveyed regions and total mean of infection was 53 percent. The samples collected from the Alfalfa field of Mashhad was propagated in the <em>Nicotiana tabacum</em> L.cv <em>Samsun</em> and then virus was purified. The Mechanical inoculation of this isolate of the alfalfa mosaic virus (ALMV) induced the local lesion in <em>Chenopodium quinoa</em>,<em>C. amaranticolor,Vigna unguiculata, Phaseolus vulgaris</em> cv <em>Redkidney</em> and the systemic vein clearing and mosaic in <em>Nicotiana glutinosa, N. tabacum</em> cv. <em>Samsun</em>, <em>Ocimum basilicum, Cicer arietinum</em> and <em>Lycopersicon esculentum</em>. In the case of the infected <em>Cucumis sativus</em>, no symptoms was observed. ALMV was purified by the method of the kaiser and Robertson(1976). The virus yield was 11/05 mg per 100g of infected tissue on the basis of serological properties.
